Alopecia areata (AA) is an autoimmune disease characterized by T cell infiltration of the hair follicle. The breakdown tolerance to follicle thought be driving force in nonscarring loss seen patients. There are currently no FDA approved treatments for AA. IL-27 plays a pleotropic role across many other diseases; however, whether its immunoregulatory effects can leveraged therapeutic applications AA unknown. To address this, we used adeno-associated virus (AAV) that drives overexpression our C3H/HeJ skin graft-induction model Briefly, AAV-IL-27 administration was followed induction using grafts from AA-affected donor. Mice were observed weekly progression. We found exogenous protected mice development. Additionally, saw increased presence FoxP3+ CD4 cells and IL-10-producing CD8 draining lymph nodes treated with AAV-IL-27, suggesting roles these populations prevention autoimmunity.
